In the drug discovery process, it is estimated that approximately 10,000 chemical entities need to be tested before one would eventually reach the consumer market. Major reasons for this high failure rate are drug toxicity, low bioavailability, biological degradation and inadvertent intrinsic side effects. Nanotechnology is exponentially expanding in the area of medicines, therapeutics, diagnostics and drug delivery. A delivery system is a formulation or a device that introduces bioactive compounds into the body and improves their efficacy and safety by controlling the rate and targeted release of the bioactive compound. Among the colloidal vectors proposed for the controlled delivery and targeting of drugs and other biologically active compounds, lipid-based nanocarriers has acquired an increasing role due to a number of peculiar technological and physical features. These carriers are generally composed of physiological lipids well tolerated by human body. The potential advantages of lipid-based nanocarriers application over other carriers are of their high biocompatibility, higher drug loading capacity and scalability. This book explores the present state of widely used lipid-based nanoparticulate delivery systems such as solid lipid nanoparticles (SLN), nanostructured lipid carriers (NLC), nanoliposomes, micelles, nanoemulsions, nanosuspensions and lipid nanotubes. The various types of lipids that can be exploited for drug delivery and their chemical composition and physicochemical characteristics are reviewed in detail. The characterization aspects and effects of their dimensions on drug delivery systems behaviour in in-vitro and in-vivo are also described. The book covers the effective utilization of these lipids based systems for controlled and targeted delivery of potential drugs/ genes for enhanced clinical efficacy. The role of lipid nanocarriers in Nanomedicine, diagnostic and therapy is also covered.
